Genomic Characterization and Evolutionary Dynamics of Developmental Gene Regulatory Networks of Paracentrotus lividus (Echinodermata: Echinoidea)
Mutations impacting DNA regulatory elements make a disproportionate contribution to evolutionary processes and are important contributors to inter-individual variation in disease susceptibility.
We are, however, limited in our ability to predict *which* mutations in non-coding, regulatory DNA are most likely to impact phenotype because DNA regulatory element sequences are often degenerate. In this study, we take a comparative approach to this problem by measuring regulatory element activity and gene expression during development in two related species. By examining the relationship between changes in DNA sequence, regulatory element activity, and expression, we will build predictive models of which kinds of mutation are most likely to impact gene expression and higher-level phenotypes. We will then test these predictions using transgenic assays.
