Interphase chromosome structure: DNA-protein and protein-protein interactions of defined active and inactive chromosomal regions of Drosophila

Differential condensation of higher order chromatin structures is essential for gene regulation in eukaryotes. The polytene interphase chromosomes are unique for studies at this level since a differential condensation of higher order structure is visible as reproducible band/interband units. On gene activation or gene silencing dynamic changes in the binding of chromatin proteins modify this basic pattern. Our groups have identified proteins specific for either condensed chromatin (bands and heterochromatin) or decondensed chromatin (interbands and/or puffs) and we have cloned large stretches of DNA from cytologically defined band/interband regions. Our objective is to define more precisely the sites and modes of binding of these proteins within the defined chromosomal intervals and to elucidate their roles played in establishing and maintaining these chromosomal structures in diploid and polytene cells. Many of these proteins function within large protein complexes. Protein-DNA, protein-RNA and protein-protein interactions responsible for chromatin binding shall be examined. We also want to find out, how this basic mode of chromatin protein-interaction is changed dynamically by processes of gene activation and silencing.
