Stereochemistry as a tool for developing anticancer agents - synthetic and functional studies on Laulimalide, a new Paclitaxel like microtubule stabilizing agent.


The part of the project performed in Prof. Saumwebers group will deal with testing the biological activity of the compounds synthesized. By indirect immunofluorescence the effect of induction and stabilization of microtubules and the induction of new microtubule organizing centres (MOC) in vivo will be assayed along with effects on the shape and integrity of nuclei and chromatin (Mooberry et al. 1999). Influence on doubling rates of the cells will be estimated by cell counts and the stage of cell cycle arrest may be determined by flow cytometry following staining by propidium iodide. Effects on nucleation and growth of microtubuli in vitro will be determined by turbidity measurements. The induction of new proteins will be followed by Western blots.
