Momentaufnahmen der photosynthetischen Wasseroxidation: simultane Röntgenspektroskopie und Kristallografie
In diesem Projekt liegt der Fokus auf der Untersuchung der gesamten Proteinstruktur und -Dynamik des Membran-gebundenen Proteinkomplexes Photosystem II (PSII) einerseits und des Sauerstoff-produzierenden Komplexes („oxygen-evolving complex” OEC) andererseits, welcher die lichtgetriebene Wasseroxidation katalysiert. Mithilfe der umfangreichen Expertise aller beteiligten Kooperations-/Projektpartner wollen wir den Mechanismus der Wasserstoffoxidation – d.h. die Oxidation von Wasser zu molekularem Sauerstoff – durch den OEC (im Detail) aufklären. Dabei dient die methodische Kopplung von Röntgen-Kristallografie und -Spektroskopie zur ganzheitlichen Beleuchtung/Untersuchung der OEC- Proteinstruktur und Veränderungen in der lokalen Umgebung (von Mn und Ca).
