SFB 1078/1: Strukturelle Grundlagen der Protonenabgabe in der Wasser-Oxidation durch das Photosystem II (TP A05)


In der oxygenen Photosynthese wird Wasser oxidiert und Di-Sauerstoff am Mn4Ca Komplex des Photosystems II (PSII) gebildet, einem 750 kDa großen Protein-Kofaktor-Komplex, der in die Thylakoidmembran von Pflanzen und Cyanobakterien eingebettet ist. Aussagekräftige PSII-Strukturmodelle sind Voraussetzung für das Verstehen von funktionell relevanten Protonierungsdynamiken. 2001 wurde das erste kristalline Strukturmodell des PSII aus dem thermophilen Cyanobakterium Thermosynechococcus elongatus veröffentlicht und in 2009 konnten die Auflösung auf 2.9 Å verbessert werden. Jüngst gelang ein Durchbruch, als die Kristallstruktur des dimeren PSII des thermophilen Cyanobacteriums T. vulcanus bei einer Auflösung von 1.9 Å erreicht werden konnte. Diese Ergebnisse liefern eine solide Basis für die Analyse der molekularen Mechanismen der Wasseroxidation. Weiterhin besteht jedoch das Problem der strahlungsinduzierten Mn-Reduktion. Wir konzentrieren uns auf die Analyse der Kopplung zwischen Elektronen- und Protonentransfer bei der photosynthetischen Wasseroxidation. Zu diesem Zweck sollen die Protonierungsdynamiken des PSII, und im Besonderen die Protonenfreisetzung aus dem wasseroxidierenden Mn4Ca Komplex, analysiert werden. Dieses erfolgt durch Röntgenkristallografie von chlorid-modifizierten Varianten und spezifischen Mutanten. Darüber hinaus soll die moderne Femtosekunden-Röntgenkristallografie bei Raumtemperatur aus PSII-Mikrokristalle angewendet werden, um die strahleninduzierte Mn-Reduktion zu überwinden sowie zeitlich hochaufgelöste Strukturdaten zu erhalten. Zudem werden die notwendigen Bedingungen für die Neutronenstreuungsanalyse erarbeitet. Die chlorid-modifizierten PSII-Varianten und die spezifischen Mutanten werden durch theoretische und spektroskopische Analysen in Zusammenarbeit mit anderen Gruppen untersucht.
Der Photosystem II-Kernkomplex (dPSIIcc) katalysiert den entscheidenden Schritt der Photosynthese – die Oxidation von Wasser. Trotz hervorragender statischer PSII-Kristallstrukturen ist der katalytische Mechanismus der Wasseroxidation am Mn4CaO5 Cluster noch nicht vollständig verstanden. Während der letzten drei Jahre haben wir entscheidende Fortschritte gemacht: wir konnten eine PSIIcc-Struktur mit einer Auflösung von 2,44 Å veröffentlichen. Diese wurde erreicht durch ein neues Protokoll zur Extraktion von Detergensmolekülen aus dem Kristall, welches zu einer neuen Kristallpackung des PSIIcc führte, die der von nativem PSII in Cyanobakterien gleicht. Wir begannen mit Femtosekunden-Röntgenstrukturkristallografischen Messungen, um die strukturellen Veränderungen am Wasser-oxidierenden Komplex („water-oxidizing complex“, WOC) im katalytischen Zyklus aufzuklären. Wassercluster-bildende Aminosäurereste des WOC wurden spezifisch modifiziert, um ein PSIIcc zu erhalten, in welchem der Übergang vom S3 zum S0 Oxidationszustand (Sauerstofffreisetzung) beeinflusst wird. Dadurch erhielten wir zunächst PSIIcc-Mikrokristalle der PsbA3 D1-Variante. Durch ein neues Microseeding-Protokoll konnte die Kristall-Uniformität verbessert und dadurch eine Auflösung von bis zu 2.5 Å erreicht werden. Durch die Bestimmung der (nah) atomar aufgelösten Kristallstruktur konnten wir Einblicke in die funktionelle Rolle des PsbO als extrinsische Untereinheit in der Protonentranslokation gewinnen. Dafür wurde eine heterolog exprimierte PsbO-Untereinheit unter Bedingungen analysiert, die es uns unter anderem erlauben, kleinste Konformationsänderungen durch die Protonenaufnahme zu erkennen. In der zweiten Förderperiode konzentrieren wir uns auf Struktur-Funktions-Beziehungen, Veränderungen des Protonierungsstatus des intakten PSIIcc sowie der der PsbO-Untereinheit, und die Rolle von einzelnen Aminosäureresten in der Protonentranslokation. Um unsere Ziele zu erreichen, planen wir die Optimierung der Auflösung von PSIIcc-Kristallvarianten mit Hilfe verbesserter Detergens-abgereicherter Kristalle. Das Mikroseeding-Protokoll für die elektronenfreie Laser-Röntgenstrahlungsmessungen (XFEL) wird weiter verbessert durch Testung der Aggregatbildung von PSIIcc unter Kristallisationsbedingungen mit Hilfe der Dynamischen Lichtstreuung. Des Weiteren wird das Makroseeding-Protokoll für die Herstellung von großen PSII-Kristallen für die Neutronendiffraktion verbessert. Außerdem wollen wir die pKa-Werte von PsbO, welche spezifisch für die Aminosäurereste sind, mit NMR messen.
