SFB 740/1: Spezifische Anreicherung von viralen Komponenten und Assemblierung von Hüllviren (TP C 03)

Nach erfolgreicher Infektion von Zellen durch Viren assemblieren sich in spezifischer und hierarchischer Art und Weise makromolekulare Strukturen, die Ausgangspunkt der Bildung und nachfolgenden Abschnürung von Viren sind. In diesem Projekt analysieren wir die Assemblierungsprozesse von Influenzaviren in vitro unter Verwendung gereinigter Proteine und Modellmembranen sowie in vivo mittels transfizierter Zellen. Die wesentlichen Fragestellungen betreffen die Wechselwirkungen des Matrixproteins M1 mit Membranen, viralen Glykoproteinen und Ribonucleopartikeln, die mit biophysikalischen Methoden untersucht werden. Langfristig werden mutierte Viren generiert, um unsere Schlussfolgerungen mit einem kompletten Modul in lebenden Zellen zu überprüfen.
